how to get the twinkle in someones eye

What sort of light would you use to get the twinkle in someones eye when shooting portraits?

The "twinkle" is called catch light. If you are taking formal portraits, the catchlight will be the result of your Main Light and, if your using one, your Supplemental light.

The important thing to remember is that if you're using two lights, you want to position them to achieve not only your desired lighting ration but also ONLY ONE CATCHLIGHT! You can see the light in your subjects eyes and move them as needed. Two obvious catchlights is a dead giveaway to articial lighting; this is often a problem seen in motion pictures short even by the greatest of cinematographers.

Hot lights, photofloods, spot lights will work; softboxes will not.
