I messed up these photos – What kind of Metering Mode / Focus (centre /multi point/ flexible) & Aperture could have yielded better results + would a low shutter speed create any difference? also would spot or centre metering have done better.

I beleive that I had not selected the Macro option.

Centre weighted average metering would have been my choice, but there doesn't seem to be too much wrong with the exposures.
Why did you use 1/30th for one and 1/500th for the other exposure? The subject isn't going to run away, you know!
The macro option is useful only with a very short distance to subject. Shorter than, say, 1 foot.

  1st (black ant nest) and 3rd (red ant nest 2) are a bit dark/underexposed. You indicate for all photos "exposure bias value -1.00". If you've inadvertently set exposure compensation to -1, that would explain the underexposure.

  Firstly thanks for your time. To answer your question Mr.smith - I was trying to check if AV or TV mode is gives me a better photo- maybe I was reading too much and had lot in my mind and ended up forgeting the basics. Jon with regards to the exposure at -1.00 I believe thats the default setting put up by the camera and I was too afraid to over expose the same by overriding it to 0 or above. Mr. smith subject around a foot away- what setting eg shutter speed and exposure mode would you two advise in future? and should the focus be centre or multi focus ?

In this case I would ideally use a tripod, fill-flash or spot metering with AEL, ISO 100 or 200, Av at F/11 or F/16, selftimer, and obviously centre focus.
