flash and camera settings

Hi, I just got a Canon EOS 5D and have a 420EX speedlite flash. Which are the best settings for an indoor event to photograph (people and speeches etc.) I usally take picture outside and I am not familiar with flash photography. Can anybody help me. What is also the best ISO to use? Thank you

A couple of helpful sites for learning how Canon's flash system works:
http://photonotes.org/articles/eos-flash/, and at the Canon Digital Learning Center there is a section called "EOS Speedlite Tips" under the "Tips and Techniques" section in the lower right.

Usually low ISO is better, but if you are not very close to your subject, then higher ISO settings extend the useful range of the flash and give more exposure to the background.
